EP219 - Α2-macroglobulin activity in endogenous uveitis in children

Purpose

To analyze α2-MG activity in children with uveitis.

Methods

The activity of α2-MG was studied in 300 samples of tears, 154 samples of serum, 39 samples of aqueous humour (AH) from 76 children with uveitis aged 3 to 17 years (mean 10.5±3.3), 135 affected eyes, in serum and tears – of 8 healthy children, in AH – of 5 somatically healthy children with congenital cataracts. The activity of α2-MG was determined by the enzymatic method with the specific substrate N-benzoyl-DL-arginine-p-nitroanilide (BAPNA).

Results

There was no correlation between the α2-MG activity in tears, serum and AH and uveitis activity and localization. There was a decrease in α2-MG activity in tears during anti-inflammatory therapy (p=0.013). There was an increase in α2-MG preoperative activity in tears in cases with severe postoperative anterior chamber (AC) inflammation (p=0.022). There was an increase in α2-MG activity in the AH with an increase in the degree of proliferative changes in the AC (p= 0.049). There was a decrease in α2-MG activity in serum with an increase in the age of patients (p=0.025).

Conclusions

An increase in the α2-MG activity in tears in the preoperative period can serve as a predictor of severe AC postoperative inflammation. AH α2-MG activity correlates with the stage of the proliferative process. Highest α2-MG serum activity is at the age of 3-6 years, with a further decrease.
